An Improved Analysis of Benzo(a)pyrene Diol Epoxide-I-protein Adduct


Abstract
Enzyme linked immunosorbent assay (ELISA) of carcinogen-protein adduct was previously developed using insoluble protease diagestion. However, the procedure of insoluble protease digestion is time consuming and expensive. In this study, acid-ethyl
acetate extraction, C18 SepPak, and ELISA with soluble protease digestion were comparatively studied. Considering recovery efficiency, sensitivity and time consumption, ELSA with soluble protease digestion was the most effective in the analysis
of
carcinogen-protein adduct. ELISA with soluble protease digestion was able to improve sensitivity and time consumption compared to insoluble protease digestion procedure. This improved ELISA could be applicable to biological human monitoring of
carcinogen-protein adduct for ancer reserch and to screening anticarcinogenic agents.
